Pefakit® Factor XIII Incorporation Assay

Pefakit® FXIII Incorporation Assay is a plasma-based functional assay for the determination of coagulation factor XIIIa activity.
FXIII substrate fibrinogen (fgen) is coated on the surface of a microtiter plate (MTP). Unspecific binding is prevented by a special blocking agent. Factor XIII in the sample is activated by thrombin and calcium ions. In the incorporation step FXIIIa in a sample incorporates the substrate 5‑biotinamidopentylamin (BAPA) into fgen coated on the MTP in the presence of calcium. The amount of incorporated BAPA is proportional to the factor XIII activity of the sample. In the next step a streptavidine-alkaline phosphatase conjugate (Strept-AP) is bound to the incorporated BAPA. Alkaline phosphatase converts the synthetic substrate p-nitro phenyl phosphate (pNPP) into phosphate and p‑nitrophenol which can be measured at 405 nm.
